• Type 2 DM
    • MR: PO 30-120mg OD
    • CR: Initially 60mg OD; can be increased by 30mg every 2 weeks. Max 120mg
    • Non-controlled: Initially 40-320mg OD/BD. Max 320mg daily.

To be taken with breakfast.

2nd generation sulfonylurea.

It binds to the β cell sulfonyl urea receptor (SUR1) thereby blocking the ATP-sensitive potassium channels consequently decreasing potassium efflux and depolarization of beta cells of the pancreas.  This causes the opening of voltage-dependent calcium channels in the beta cells resulting in calmodulin activation and the consequent exocytosis of insulin-containing secretory granules.
