• Type 2 DM:
    • Initial dose: PO 2.5mg/500mg OD/ BD
    • Max: 20mg/2000mg daily, divided into multiple doses.

Tablet:

  • 2.5mg/500mg
  • 5mg/500mg
  • Take with meals to reduce gastrointestinal side effects

Glibenclamide: Sulfonylurea. It stimulates insulin secretion from functioning pancreatic beta cells, which lowers blood glucose levels.

Metformin: Biguanide. It decreases hepatic glucose production, reduces intestinal glucose absorption, and improves insulin sensitivity by increasing peripheral glucose uptake and utilisation.

  • Hypoglycemia
  • Weight gain
  • Gastrointestinal disturbances (nausea, vomiting, diarrhoea)
  • Metallic taste

Black Box warning:

Risk of lactic acidosis due to metformin accumulation, especially in patients with renal impairment, liver disease, or conditions that increase lactate production.
